Hermann II. folgte 1122 seinem Vater als Graf von Winzenburg, wurde 1123 Markgraf von Meißen und 1124 Landgraf von Thüringen, ohne jedoch die beherrschende Stellung seiner Nachfolger zu haben. Er war jahrelang im engsten Gefolge von Erzbischof Adalbert I. von Saarbrücken-Mainz, zeigte sich jahrelang als haltloser Fehdetyp und ließ 1130 den Grafen Burchard von Locchum-Friesland, einen engen Vertrauten Kaiser LOTHARS III., von seinen Leuten erschlagen. Deshalb wurde er auf dem Fürstentag zu Quedlinburg vom 18.8.1130 verurteilt und alle seine Würden und Lehen wurden ihm entzogen. Hermann mußte sich Kaiser LOTHAR III. als Gefangener überliefern, weilte ab 1130 jahrelang im Rheinland im Exil und war möglicherweise zeitweise Kommandant von Segeberg. Er tauchte ab 1138 als Anhänger KONRADS III. wieder auf und wurde zeitweise Graf von Plesse genannt. Er war mainzischer Vasall, Gegner der WELFEN und NORTHEIMER im Thronkrieg, bekriegte beide und söhnte sich 1140 mit letzteren aus und bahnte damit den Erbanfall von 1144 an. Er schloß sich wieder eng an KONRAD III. an und wurde dessen Schwager. Er galt seitdem als Reichsfürst, tauchte stets im engsten königlichen Gefolge auf und war Zeuge in vielen königlichen Urkunden. Er stritt ständig mit den Bischöfen von Halberstadt und den Äbten von Corvey wegen vorenthaltener Lehen und wurde am 8.5.1150 wieder mit der 1130 bei seiner Verurteilung verlorenen hildesheimischen Lehenburg Winzenburg belehnt. Er beerbte 1146 seinen Bruder Heinrich, besaß damit einen bedeutenden Machtkomplex zwischen Hameln, Hildesheim, Wolfenbüttel, Göttingen, Goslar und Einbeck und war den WELFEN in Sachsen an Macht und Einfluß fast ebenbürtig. Er nahm 1147 am erfolglosen "Wendenkreuzzug" Heinrichs des Löwen teil. In der Nacht vom 29. zum 30. Januar 1152 wurde er mit seiner schwangeren dritten Gemahlin auf seiner Burg Winzenburg von der Burgbesatzung eventuell auf Veranlassung des Bischofs von Hildesheim, erschlagen. Sein Besitz fiel nach jahrelangem Erbkrieg an Heinrich den Löwen.
Hermann wurde von seinem Nachbarn, dem Grafen Heinrich von Bodenburg, ermordet.

Hermann II succeeded his father as Count of Winzenburg in 1122, became Margrave of Meissen in 1123 and Landgrave of Thuringia in 1124, but without having the dominant position of his successors. For years he was in the closest entourage of Archbishop Adalbert I of Saarbrücken-Mainz, showed himself to be an unstoppable feud type for years and in 1130 had Count Burchard of Locchum-Friesland, a close confidant of Emperor Lothar III, killed by his people. For this reason he was sentenced at the Quedlinburg Princes' Day on August 18, 1130 and all his dignities and fiefs were withdrawn from him. Hermann had to ask Emperor LOTHAR III. handed down as a prisoner, lived in exile in the Rhineland for years from 1130 and was possibly commander of Segeberg for a time. He appeared from 1138 as a follower of CONRAD III. again and was temporarily called Count von Plesse. He was a vassal of Mainz, an opponent of the WELFEN and NORTHEIMER in the War of the Throne, fought both and was reconciled with the latter in 1140, paving the way for the inheritance of 1144. He again closely aligned himself with KONRAD III. and became his brother-in-law. Since then he has been regarded as an imperial prince, has always appeared in the closest royal entourage and has been a witness in many royal documents. He constantly quarreled with the bishops of Halberstadt and the abbots of Corvey because of withheld fiefs and was again enfeoffed on May 8, 1150 with the Hildesheim fiefdom Winzenburg, which had been lost when he was sentenced in 1130. He inherited his brother Heinrich in 1146, thus possessing a significant power complex between Hamelin, Hildesheim, Wolfenbüttel, Göttingen, Goslar and Einbeck and was almost equal in power and influence to the WELFEN in Saxony. He took part in Henry the Lion's unsuccessful "Wend Crusade" in 1147. In the night from January 29th to 30th, 1152, he was killed with his pregnant third wife at his castle in Winzenburg by the castle garrison, possibly at the instigation of the bishop of Hildesheim. After years of inheritance wars, his possessions fell to Henry the Lion.
Hermann was murdered by his neighbor, Count Heinrich von Bodenburg.
