Sir Hugh was the son of Thomas Despencer, and brother to both Thomas, who died before October 1218. He was a descendant of a noble family who came to England from the same location as William the Conqueror and at the same time. This family, surnamed De Albetot, became William's Stewards. Sir Hugh was appointed High Sheriff of Staffordshire and Shropshire in 1222 and High Sheriff of Berkshire in 1226 and 1238.

Sir Hugh le Despenser, feudal Lord of Loughborough, Burton, Hugglescote, Freeby and Arnesby , Leicestershire, Parlington and Hillam, Yorkshire, Sibsey and Aukborough, Lincolnshire and R yhall and Belmesthorp, Rutland. [Burke's Peerage]King Henry III, in the 8th year of his reig n [1224], constituted Hugh de Spencer sheriff of the counties of Salop and Stafford, and gove rnor of the castles of Salop and Bruges (Bridgnorth). He was subsequently sheriff of Berkshir e and governor of Wallingford Castle. To this Hugh Henry II gave the manor of Rithal, co. Rut land, and in the 21st of that monarch's reign, upon the death of John Scot, Earl of Chester , he was deputed with Stephen de Segrave and Henry de Aldithley to take charge of the castle s of Chester and Beeston. After this Hugh came his grandson, another Hugh Despencer. The king has granted to Hugh Despenser a moiety of the manor of Chalgrove with the capital messuage, and to Hugh de Plessetis, Drogo de Barentin and Nicholas de Boleville the other moiety with appurtenances formerly of Peter de Maulnay, in order to sustain them in the king's service for as long as it pleases the king. Order to the keeper of the honour of Wallingford that, bythe view and testimony of trustworthy and law-worthy men of his bailiwick, he is to cause Hugh Despenser to have full seisin of a moiety ofthe same manor with the aforesaid capital messuage, and the abovesaidHugh, Drogo and Nicholas to have full seisin of the other moiety of the aforesaid manor with appurtenances, saving to Alexander of Dorset the corn that he bought from Peter's bailiffs from the land that Peter had in Chalgrove, and saving to the king the rest of the same corn with the rest of the stock and chattels found in the same manor.

Henry III King of England granted custody of land of "Johannis de Sancto Johanne", during the minority of his heirs, to "Galfrido Dispensario" and granted the marriage of "dictorum heredum" to "Hugoni Dispensario fratri ipsius Galfridi" dated [Apr] 1230[151]. Note: Sir Hugh le Despenser (died 1238) was a wealthy landowner in the East Midlands of England, and served as High Sheriff of Berkshire.
He was probably the son of a Thomas Despencer, and brother to both Thomas, who died before October 1218, and Rohaise, who married Stephen de Segrave. It is said that he married Mary de Quincy, daughter of Saer de Quincy (died 1219) and Margaret de Beaumont (died 1234). Hugh and Mary probably had at least three children: Pernell, who married Geoffrey Savage; Mary, who married Roger St. John; and Hugh le Despencer, 1st Baron le Despencer (died 1265).
Sir Hugh was appointed High Sheriff of Staffordshire and Shropshire in 1222 and High Sheriff of Berkshire in 1226 and 1238.
He held eleven manors in England: in Leicestershire, Yorkshire, Lincolnshire, and Rutland. He is reported to have been instrumental in the repairs of 1232 to Porchester Castle in Hampshire, a site first used by third-century Romans. Sir Hugh had this ever-evolving castle updated to include a new forebuilding to the keep and portcullises for the gatehouses, and completed repairs to the wall and hall.
He is believed to have married Mary de Quincy, a daughter of Saer de Quincy, 1st Earl of Winchester (d.1219) by his wife Margaret de Beaumont
